Th1 and Th2 Cytokine Expression in Allergic mucosa of the Nasal Cavity



Abstract
In order to assess the role of cytokines in the pathophysiology of allergic rhinitis we observed the patterns of expression of cytokine mRNA in the nasal mucosa of 10 patients with perennial allergic rhinitis and 10 control subjects using reverse
transcription-polymerase chain reaction (RT-PCR) for Th2 cytokine (IL-4, IL-5) and Th1 cytokine (IFN-¥ã).
Expression of IL-4 and IL-5 mRNA's detected by Southern blot of RT-PCR products was more often observed in patients with allergic rhinits than in control subjects. When the level of expression of cytokines and ¥â-actin mRNA was measured, the mean
density ratios for IL-4 and IL-5 were higher in patients with allergic rhinitis than in control sujbects, while there was no significant difference in the mean density ratio for IFN-¥ã between the two groups. This study demonstrates that both
IL-4
and
IL-5 m-RNA's are expressed significantly in the nasal mucosa of patients with perennial allergic rhinitis and these results suggest that Th2 cytokines may play an important role in the development of nasal allergy.
